چکیده

Background Diffusion MRI is a valuable tool to assess white matter (WM) microstructure biomarkers of brain aging and Alzheimer’s disease (AD). The most common model used extract diffusion properties the tensor (DTI). However, DTI cannot resolve complex WM suffers from partial volume effects. Further, majority multi-site imaging clinical studies, such as ADNI, are limited single-shell acquisitions due time constraints. Therefore, multi-compartment models not feasible overcome these limitations except multi-tensor models, Ball-and-Stick (B&S) under some B&S offers novel approach derive for early detection AD because it splits diffusion-weighted MR signal into several anisotropic components (Stick) single isotropic component (Ball). This implemented in Anima software. Here, we propose classification using four two indices quantified various anatomical regions interest (ROIs) compare their ability distinguish AD. Method Cross-sectional data were available 413 ADNI-3 participants: 68 (age: 76.81±8.3; 50% women) 345 cognitively normal (CN; age:72.91±7.27; 62% women). We processed T1-weighted sequences following pipeline Fig.1. obtained maps: fractional anisotropy (FA), axial (AxD), mean (MD), radial (RD) diffusivity; free water weight (FWW) Stick-axial diffusivity (ST). maps within set ROIs. Then, performed high-dimensional pattern approach. For each metric, consider all ROIs jointly identify minimal whose values maximally differentiate between CN. Result with regional FWW ST measures shows greatest discrimination CN (FWW ROCAUC = 0.942; 0.938) compared (Table 1). Left PHG Parahippocampal Gyrus Right Hippocampus have highest contribution 2, Fig.2). Conclusion Our results suggest more sensitive discriminate than measures, widely research. has advantage identifying separating crossing fibers, therefore, potential offer greater insights underlying pathology

Diffusion MRI of Brain Connectivity and Microstructure:

What is a “diffusion coefficient” or “diffusion tensor” in living tissue, anyway? While the measurement of the self-diffusivity in an NMR tube represents a "gold standard", in complex media particularly in living tissue, water “diffusivity” ceases to have a clear meaning or definition. The apparent diffusion coefficient (ADC) and apparent diffusion tensor (ADT) concepts were introduced to answe...

Model-based super-resolution of diffusion MRI for microstructure imaging

PURPOSE This work develops a super-resolution reconstruction (SRR) technique that constructs isotropic high-resolution (HR) diffusion-weighted images (DWI) from multiple anisotropic lowresolution (LR) acquisitions (Fig 1). Isotropic HR DWIs enable the mapping of tissue microstructure for fine brain structures, such as the subfields of hippocampus. Direct acquisition of such images require prohi...

The Participation of Three Brain Tissues in Alzheimer’s disease Diagnosis from Structural MRI

Alzheimer’s disease (AD) is a progressive and irreversible disease which gradually makes patients unable to do their daily routines. Although the present treatments can not cure the disease completely, its early detection can reduce symptoms and enhance the patients’ life quality. In the current literature, using the grey matter (GM) tissue which is known as an appropriate biomarker is highly c...

Dissociable diffusion MRI patterns of white matter microstructure and connectivity in Alzheimer’s disease spectrum

Recent efforts using diffusion tensor imaging (DTI) have documented white matter (WM) alterations in Alzheimer's disease (AD). The full potential of whole-brain DTI, however, has not been fully exploited as studies have focused on individual microstructural indices independently. In patients with AD (n = 79), mild (MCI, n = 55) and subjective (SCI, n = 30) cognitive impairment, we applied linke...

Evaluating the Accuracy of Diffusion MRI Models in White Matter

Models of diffusion MRI within a voxel are useful for making inferences about the properties of the tissue and inferring fiber orientation distribution used by tractography algorithms. A useful model must fit the data accurately. However, evaluations of model-accuracy of commonly used models have not been published before. Here, we evaluate model-accuracy of the two main classes of diffusion MR...
